Knight joins the JJ Abrams-produced adaptation of Stephen King's best-selling novel as Johnny Clayton, reports TVLine.
James Franco stars in the Hulu project as Jake Epping, a high school teacher who travels back in time in an attempt to stop the assassination of US president John F Kennedy.
Knight's character Clayton is a Texas salesman whose wife Sadie (Sarah Gadon) falls for Jake.
Daniel Webber, Chris Cooper and Cherry Jones are among the other stars who will appear in the series.
King's novel is being adapted for television by Friday Night Lights' Bridget Carpenter.
Aside from playing George O'Malley in Grey's Anatomy, Knight recurred in The Good Wife and featured in baseball movie 42.
